After I recently put a new fan shroud, hoses, thermostat, my car spits up a little coolant through the run-off hose after I run it for a couple minutes. It is a small puddle size on the ground. Did I do something wrong?

Yes. Your "run-off hose" is supposed to be routed/connected to an overflow bag/bottle. The overflow bag/bottle has a "run-off hose"connected to it, but not the radiator. The pressurized cooling system will use the overflow bag/bottle to temporarily store expanding and contracting coolant.

So, the run-off hose is connected right below the radiator cap right? Itgoes to the overflow bottle and since it is pressurized, will the fluid return to the radiator,or do I need to manually dump it back in the radiator or leave it sitting there?
early mustangs didnt have over flow bottles. as the car heats up, coolant expands and some has to escape...hence why jcthorne said there needs to be a space at the top of the radiator to allow for this. its normal and you can live with it. you just need to check your coolant levels regularly...which you should do anyway whether or not you have an overflow bottle. If all is healthy, you probably only have to top occaisionally.
if you go down davesanborns path, you can add an overflow like a modern car. coolant from expansion collected in the bottle and sucked back into the radiator as the car cools and the pressure drops.
BUT, you must have a return style radiator cap. The original ones are one way. The return cap has an additional rubber seal thingy which allows coolant to be drawn back in.

the leak is from the overflow hose. over time it will fix itself. these engines will fix themself to where they like to run. the water level will settle down, and it may look like its low, but its most likely not. its just where the engine likes it to be at.
my dad just installed a nice looking overfill tank in his 66. its pretty fancy.
